Abstract The patient after surgical need a therapeutic period that requires brief-term hospitalization in the daycare unit, and they are usually immunocompromised that can easily have a nosocomial infection. The environment and equipment surround the patient can be a factor of transmission. This study investigates the presence of bacteria by isolate and identifies them by microbial, biochemical, and molecular techniques. 60 swab samples were isolated from the daycare unit in king Abdulaziz university hospital. Samples collected from equipment's surface (bathroom, bed, table, fridge, chair, closet, wall, Iv pump, Sphygmomanometer) and nurses' hands. The results indicated that bacilli and (8) cocci were based on cultural, morphological and biochemical characteristics (60) of bacterial isolates. There were also Gram-positive isolates (67) and Gram-negative isolates (1). Five (5) generic ware isolates are based on 16S rRNA gene sequencing techniques, namely Bacillus, Staphylococcus, Microbacterium, Escherichia coli, and Bhargava. The most occurring bacteria belonged to Bacillus subtilis. Overall, the present study indicated that high touch surfaces are habitually contaminated with pathogens that cased infection to the patient, and it is important to focus the cleaning of these sites and improve the cleaning protocol to controlling the nosocomial infection.
